Embryonic stem (ES) cells can differentiate into derivatives of all three of the primary germ-cell layers, including cardiomyocytes, and previous studies have suggested that early steps in embryonic cardiomyogenesis take place during embryoid body (EB) differentiation of ES cells

Our results shed light on the role of sik1 in cardiomyogenesis and, although further investigation is required to solve this issue, our data provide a new piece of puzzle in the understanding of the molecular mechanisms regulating cardiomyocyte cell cycle, which is of vital importance for future clinical approach.

Collectively, we provided evidence that sik1-mediated effects are specific for cardiomyogenesis regulating cardiomyoblast cell cycle exit toward terminal differentiation.

Taken together, these data indicate that the sik1flp/flp population can differentiate into contracting cardiomyocytes, but showing a consistent delay in cardiomyogenesis. sik1 led to a delayed differentiation, we assessed the proliferative potential of sik1flp/flp ES cells by BrdU incorporation, postulating that the lack of sik1 might increase the proliferation rate of these cells.
