Human IgG (Baxter Heathcare Corporation, Westlake Village, CA) or murine IgG (Jackson ImmunoResearch Laboratories, Inc.) were added to the oxidized NP suspension in PBS with 50 mM of sodium cyanoborohydride in 1 M NaOH at final concentrations of IgG and SPIO of 2 mg / ml and 10 mg / ml, respectively.
This labile intermediate was immediately reduced with sodium cyanoborohydride in order to stabilize the bond between the protein and the NP.
Sodium periodate, sodium cyanoborohydride, neocuproine, ammonium acetate, ascorbic acid and potassium permanganate were purchased from Sigma-Aldrich, St. Louis, MO.
