Methylumbelliferone (MU; ε at pH 7.0) derived from MU-Man hydrolysis was measured continuously by a fluorimeter with an excitation wavelength of 365 nm and an emission wavelength of 440 nm.
Measurements were done at 37°C with constant stirring in acrylic cuvettes using a Fluorolog fluorimeter.
The patches are monitored by the coaster, which is a miniature fluorimeter, able to measure either excitation ratio for CO2 and pH or fluorescence lifetime for O2.
Fig 6 at 37°C, washed twice with PBS and red reporter fluorescence was estimated in a microplate fluorimeter (ThermoFisher).
The fluoroSENS is a versatile bench-top fluorimeter with single photon counting sensitivity and an excellent water Raman signal-to-noise ratio.
